Research Abstract

1.牛血清のサンプリング
ネオスポラ原虫抗体陽性牛のうち、流産あるいは異常産を起こした母牛および正常な出産をした母牛の血清サンプリングを実施した。当初、山形県家畜保健衛生所を介したサンプリングを予定していたがサンプル数の確保が困難だったため、宗谷家畜衛生保健所よりサンプルを確保した。
2.原虫由来タンパク質と組換えタンパク質の回収
プロテオームマップの作成およびELISA法の抗原に用いるために、分離精製したネオスポラ原虫NC-1株(犬から分離した世界的標準株)から可溶性タンパク質を調整した。またELISA法に用いる原虫由来組換えタンパク質として、大腸菌発現系によるNcSAG1,NcSRS2,NcGRA7の生産・精製方法を確立した。
3.原虫由来組換えタンパク質を用いたELISAによる牛血清のスクリーニング
上記組換えタンパク質を抗原としたELISA法により牛血清のスクリーニングを実施したところ、ネオスポラ陽性で正常出産した群と比較して、ネオスポラが感染し流産を起こした個体において抗NcGRA7抗体の上昇が確認できた。
4.二次元電気泳動による虫体分泌タンパク質のマッピング
回収した原虫タンパク質液を二次元電気泳動にて展開した後にタンパク質染色(クマシー染色・銀染色など)を行い、さらに画像解析を行うことでプロテオームマップを作成した。
5.各分泌タンパク質のMALDI-TOF-MS分析
回収した原虫タンパク質液を二次元電気泳動にて展開した後に、スクリーニングした牛血清を用いたイムノブロッテイングを行い、流産あるいは異常産を起こした母牛および正常な出産をした母牛の血清が認識する原虫由来分子を比較検証することを予定している。
