Research Abstract

結合組織成長因子(CTGF)は90年代初頭に内皮細胞培養上清中に、その存在が確認されたばかりの新しい成長因子である。その働きは内皮細胞、上皮細胞、神経細胞などいくつかの細胞種に作用し、血管形成、動脈硬化、神経痂皮形成等にも関与する多様な生物学的作用を持っている。中でも血管新生作用はVEGFと比較して遜色ない作用を有している事が判明した為、CTGFの遺伝子発現制御機構、特に腫瘍血管新生時における制御機構と解明する事は、より効果的な抗血管新生療法を開発する糸口を提示すると思われた。よって研究代表者は腫瘍血管新生時の癌組織の微少環境である低酸素状態を模倣した実験的低酸素下のCTGF遺伝子発現制御機構の解明を目的としている。研究代表者らはこれまでにctgf mRNAの3'側非翻訳領域(3'-UTR)に遺伝子発現抑制性elementを発見し、当該84bpの領域に細胞特定因子が結合することで低酸素下のctgf mRNAの安定性を増大させていることを報告した(Kubota, et. al. Oncogene2000,Kondo, et. al. Oncogene2006)。現在までに,その細胞特定因子の同定は終了し当該RNAとの結合の再確認も複数の手法で既に確認した。現在同定したタンパクのRNAへの結合に関する責任領域の絞り込みを行いN末にその責任領域が存在する傍証を掴んだ。さらに絞り込みを行っている段階である。
