Research Abstract

本年度は,アルツハイマー病関連遺伝子カイコオルソログ,14-3-3epsion(14E),zeta(14Z)について機能解析を行った。はじめに,絹糸腺を二次元電気泳動で分離し,Xpress-tagged14-3-3plobeと反応するスポットについて解析したところ,カイコHeat Shock Protein 60(BmHSP60)であることが明らかとなった。HEK293細胞に14Eまたは,14ZとBmHSP60を共発現させ,免疫沈降法により,両者の結合を確認することができた。さらに,BmHSP60と14Eの結合部位を探索するために,BmHSP60のN末端部位(2-261)およびC末端部位(262-572)の組み換えタンパク質を作製し,同様の方法で両者の結合を検討した。その結果14Eは,BmHSP60のN末端部分とリン酸化非依存的に結合することが明らかとなった。次に,カイコ発生ステージおよび5齢幼虫期間の主要な組織における,14E,14Z,BmHSP60の発現をウエスタンブロティングで検討した。14E,14Z,BmHSP60ともに全発生ステージの主要な組織で高発現していることが明らかとなった。
14-3-3は細胞シグナル伝達を担う分子アダプターである。一方Hsp60はタンパク質フォールディングを制御する分子シャペロンである。両者の結合は,カイコ細胞におけるタンパク質成熟と機能発現の連携に重要な役割を果たしていると思われた。この研究成果は,Neuroscience Researchに投稿した(印刷中)。
