Outline of Annual Research Achievements

本研究の目的は、希少がんであるヒトの神経内分泌腫瘍(neuroendocrine tumor; 以下NET)の症例解析とモデルマウスの作製・解析を通じてNETの本態を解明することである。
NETは、神経内分泌細胞に由来する腫瘍であり、膵臓、下垂体、甲状腺、消化管、肺など全身の様々な臓器に生じる。希少がん故にサンプルの収集が難しいことやマウスモデルの開発が遅れていることにより、未だ腫瘍発生機序は十分に解明されていない。我々は、p53標的遺伝子であるPHLDA3遺伝子が、がん遺伝子Aktの抑制因子をコードし、肺と膵臓のNETにおいて、がん抑制遺伝子として機能している事を明らかにした(Cell, 2009、PNAS, 2014)。我々は、PHLDA3が様々な臓器に由来するNET共通のがん抑制遺伝子であると考えており、本研究課題を進めることで、臓器を超えたNET共通のがん抑制メカニズムを解明し、NET研究とそれに続く創薬や治療法開発につなげたい。
