Outline of Annual Research Achievements

(i)Bradyrhizobium属根粒菌の共生アイランドの動的進化の解明
前年度sacB選抜とRj2ダイズ選抜の欠失パターンが異なっていた。sacB特異的な欠失株をRj2ダイズに接種を行ったところ、(競合的)根粒形成能が明らかに低下していた。したがって、Bradyrhizobium属根粒菌の共生アイランドは自由生活(培養過程)で確率的に挿入配列(IS)介在型欠失が生成しており、Rj2ダイズ選抜ではその一部が選択されていることが判明した。さらに、根粒菌コレクションから共生アイランド内のIS間が重複している獲得型株が見出された。以上の結果は、根粒菌の共生アイランドが挿入配列(IS)を介して欠失と重複を起こす動的な存在であることを実験室内で証明したものとして、高く評価できる。
(ii) Bradyrhizobium属の窒素固定エンドファイトと根粒菌の共有進化
ダイズ根粒から分離したBradyrhizobium属根粒菌を、レニーの半流動培地に接種し、アセチレン還元活性を測定したところ、自由生活で窒素固定能を発現する非共生型Bradyrhizobium属細菌と同じレベルでアセチレン還元活性を示す株は得られなかった。したがって、本アプローチからは共生アイランドとゲノムコアの両方にnif遺伝子群を持つハイブリッド型のBradyrhizobium属細菌は見出されなかった。ソルガム根由来のBradyrhizobium属細菌は「根粒菌型」、「自由生活窒素固定型」、「非窒素固定型」3つの窒素固定様式に分類された。それらの詳細な系統解析とゲノム解析により、ある系統において「根粒菌型」と「非窒素固定型」の両方のタイプが共存し、Bradyrhizobium属細菌において、共生アイランドの水平伝達が起りやすい系統の存在が示唆された。
