Outline of Final Research Achievements

Xyloglucan/xyloglucan-oligosaccharide degradation-related enzymes, such as isoprimeverose-producing oligoxyloglucan hydrolase, β-galactosidase, extracellular- and intracellular-α-xylosidases and xyloglucan-specific endo-β-1,4-glucanases, were identified and characterized in Aspergillus oryzae. The gene expressions of some of these glycoside hydrolases were induced in the presence of xyloglucan-oligosaccharides and these enzymes hydrolyzed xyloglucan/xyloglucan-oligosaccharides cooperatively.

Academic Significance and Societal Importance of the Research Achievements

麹菌は醸造産業において重要な微生物であり、多糖類やタンパク質を分解するために様々な酵素を生産している。キシログルカンは植物の細胞壁や種子に含まれている複雑な側鎖構造を有する多糖類であり、本研究では、麹菌がこのキシログルカンやキシログルカン由来オリゴ糖を分解する酵素群を明らかにした。麹菌はこれらの酵素を分解対象の基質やその分解物が存在する場合に積極的に生産し、これらの酵素を協調的に基質に作用させることによって巧みにキシログルカンを分解している。このことは、麹菌にはまだ認知されていない分解能力(酵素)とそれを引き出すためのきっかけ(誘導物質)があることを示唆している。
