Outline of Annual Research Achievements

トンボは、基本的に視覚で相手を認識するため、体色や斑紋に著しい多様性が見られる。申請者らは、色覚に関わるオプシン遺伝子がトンボで極端に多様化していること、オプシン遺伝子の発現は幼虫と成虫で大きく異なることを見出した。また、アカトンボの体色変化は色素の酸化還元反応が原因であること、シオカラトンボの体色変化はUV反射Waxの産生によることを発見した。これらの知見により「トンボの幼虫と成虫で色覚はどのように変化するのだろうか？」「トンボの体色変化には、どのような遺伝子が関与するのだろうか？」などの素朴な疑問が浮かび上がった。これらの疑問の解明に向けて、申請者らはトンボの遺伝子機能解析系と実験室内飼育系を確立することに成功した。本研究課題では、申請者らが構築した一連の実験系を用いてトンボの色覚と体色形成の分子基盤を深いレベルで解明することを目指す。2022年度は、成虫の体色形成に関わる遺伝子のうち、メラニン合成経路に関与する遺伝子群に関してトンボと他の昆虫の対応関係を整理し、筆頭責任著者として論文発表を行うことができた。また、トンボを中心とした昆虫の性分化に関するこれまでの知見を整理して総説を発表した。また、昆虫の体色について日本語で整理し、「動物の体色がわかる図鑑（グラフィック社）」を共著で刊行することができた。さらに、前年度に引き続き、色素合成や模様特異的に発現する遺伝子の機能解析を行い、体色に関わる新規遺伝子を複数同定することに成功した。

Reason

体色に関わる遺伝子の探索および機能解析は順調に進んでおり、トンボのメラニン合成酵素や性分化に関して論文発表を行うことができた。なお、一部のサンプルは、入手が難しいものがあったため、研究期間の延長を行って対応している。

Strategy for Future Research Activity

これまでに体色と関連がみられた遺伝子を多数同定することに成功したため、RNAiによる機能解析およびRNAi後のRNAseq解析によって引き続き色素合成やパターン形成に関わる遺伝子の解明を進めていきたい。
