Outline of Annual Research Achievements

摂食により腸内分泌細胞から分泌されるインクレチンはグルコース濃度（血糖）依存性にインスリン分泌を増強する。この作用を利用して開発されたインクレチン関連薬は現在糖尿病治療薬として広く使用されているが、一部の糖尿病や肥満症で応答不全（ノンリスポンダー）が認められており、その機序は不明である。本研究では、1) 糖尿病におけるインクレチン応答性インスリン分泌不全の機序を解明し、2) それを標的とした治療戦略を確立することを目的とする。2019年度は各項目においてそれぞれ以下の研究を行った。
1) インクレチン応答性の低下を示す肥満糖尿病モデルZFDMラットから単離した膵島のトランスクリプトームおよびメタボローム解析を行った。特に肥大した膵島においてβ細胞の脱分化や解糖系の亢進、ミトコンドリア機能の低下など、腫瘍細胞様の代謝変化が認められ、インクレチン応答不全を含む病態形成に寄与していることが示唆された。また昨年度に引き続き、ZFDMラットの膵島で認められるO-Nアセチルグルコサミン（O-GlcNAc）化の亢進とそのインクレチン応答不全における関与を明らかにするために、O-GlcNAc修飾を変化させた膵β細胞株を用いてプロテオーム解析を行った。
膵β細胞特異的KATPチャネル欠損マウスの解析から、高血糖などで持続的に脱分極したβ細胞では細胞内シグナル伝達が変化することでGLP-1とGIPに対する応答性に差異が生じることが明らかになった。
2) 糖尿病治療薬であるスルホニル尿素薬とcAMPおよび、cAMPに関連するチャネルのリガンドをクエリとしたインシリコ類似化合物検索およびインスリン分泌アッセイによるスクリーニングにて得られた候補化合物について、前年度に引き続き構造活性相関解析を行い、さらに細胞内標的探索を目的とするケミカルプロテオミクスのためのタグ付加部位の検討を行った。

Strategy for Future Research Activity

1)糖尿病におけるインクレチン応答性インスリン分泌不全の分子機構の解明
今年度は膵β細胞株のプロテオーム解析で得られたデータを解析し、インクレチン応答不全を中心としたインスリン分泌障害に関与するO-GlcNAc修飾タンパク質の候補を絞る。得られた候補分子について、ノックダウンややノックアウト細胞株の作製などにより機能解析を行い、インクレチン応答性インスリン分泌における役割を明らかにする。
2)インクレチン応答性インスリン分泌不全を標的とした新たな治療戦略の確立
化合物の標的探索のためのケミカルプロテオミクスを引き続き行う。タグ付加した化合物の効果を再度評価した後、プルダウン法により化合物と標的タンパク質の複合体を精製し、プロテオーム解析を行う。得られたデータの解析により化合物の標的分子を同定する。同定された標的分子について、機能解析を行いインスリン分泌における役割を明らかにする。また、前年度までに得られた高活性化合物について、インスリン分泌特性や病態モデル動物における効果をより詳細に解析し、有効性を明らかにする。
