Outline of Annual Research Achievements

生命システムの頑健性は、複雑な形態の進化がどのようにして起こってきたかを解明するための重要な概念の一つである。本年度はウニの遺伝子制御ネットワークの頑健性が、胚発生過程における遺伝子発現プロファイルのばらつきをどの程度許容するかについて検討した。ウニの３ペアの雌雄を用い、人工授精を行い、それぞれから発生する胚を用いて遺伝子発現プロファイルを得た。その３検体（バッチ）を比較して、検体間でどの程度遺伝子の発現にばらつきが見られるか調査した。ここでは、遺伝子制御ネットワークに焦点を当てるため、転写因子に対象を絞った。その結果、573の転写因子のうち、17の遺伝子が3つのバッチのうち1つのバッチの卵にのみ母性ＲＮＡとして蓄えられていることがわかった。また、母性発現しておらず、調べた初期発生の段階でzygoticな発現を示す転写因子94遺伝子のうち、4遺伝子は1つのバッチでのみ発現を示し、14遺伝子は2時間以上の発現開始時間の違いを示した。いくつかの細胞系列における遺伝子発現を調べることで、バッチ間で遺伝子発現開始の相対的なタイミングにばらつきがあることを検出した。観察された個体間のばらつきが、バッチ間のゲノム配列のばらつきのみによるものなのか、卵に蓄積するRNAの違いによるものなのかは定かではないが、海産動物の自然個体群では、胚発生が頑健であり、多様な遺伝子発現プロファイルを許容しながら発生が進行していることを示す証拠が得られた。このような多様な遺伝子発現を許容することで、新しい発生プロセスを探索する自由度が上がり、新しい形態の進化をもたらす潜在能力につながっていると考えられる。集団発生生物学という新しい研究分野が生まれつつある。
