| Outline of Annual Research Achievements |
外分泌のメカニズム解明を目指し、シェーグレン症候群モデルとして使用されるnon-obese diabetes (NOD) マウスを用いて涙腺・唾液腺における形態の観察および分子生物学的検討を行った。NODマウスの雄は涙腺炎を、雌は唾液腺炎を高確率で発症するため、実験には性周期に影響されない雄マウスを用いた。
涙腺炎発症後の雄のNODマウスの涙腺において、対照マウスの涙腺と比して肥大していることが判明し、副交感神経刺激後の涙液分泌量が低下していた。一方で同一マウスにおいて、唾液分泌量の減少は認められなかった。涙腺において、水分泌に関与するaquaporin 5 (AQP5) が減少していたが、タンパク分泌に関与するmyristoylated alanine-rich C kinase substrate (MARCKS) は変化していなかった。また、肥大化に関与するyes-associated protein (YAP) は変化していなかった。以上より、涙腺炎発症後の雄のNODマウスの涙腺において、AQP5の減少が涙液分泌低下に関与する可能性が考えられた。
なお、2018年度に科研費・研究活動スタート支援で始めた本研究は、2019年度より科研費・若手研究に切り替え継続する。外分泌低下を発症する前後を比較し、外分泌低下に関与する因子を同定する予定である。
