Outline of Annual Research Achievements |
The goal of my research was to fabricate bioartificial pancreatic islets and optimise their culture conditions. The first key point of my work is the use of oxygen permeable PDMS-chip to fabricate pancreatic spherical aggregates (spheroids), which provided cells with adequate oxygenation and prevented development of hypoxic core during long culture (1 week). The next key point is the use of the antioxidant ascorbic acid-2-phosphate in culture medium, which protects cells from reactive oxygen species generated due to the fact that in vitro conditions differ from the optimal in vivo ones. The third key point is the glucose concentration in culture medium. Although monolayer is culture in high glucose, my investigation showed that lower glucose concentration is beneficial for spheroid culture.
