Outline of Annual Research Achievements

再生医療の治療効果向上に向け,血管構造を有し不適切な細胞が除去された活性の高い3次元細胞組織の生成が求められている.本研究では,血管構造の効率的な形成に向けた細胞のパターニング技術,不適切な細胞の除去技術,3次元化に資する細胞シートの回収技術を超音波を用いて確立した.上記の技術は,超音波による音響放射力,および音響流を細胞に付与することで達成可能なものである.これらの成果は3報の原著論文に掲載されている.さらに,現在一報の原著論文が査読中である.これらに限らず、本研究は多くの関連研究を生み出した.
細胞のパターニングに関しては,汎用的な細胞培養用のディッシュ内に超音波を照射することで圧力勾配を発生させ,圧力の低い位置に細胞を集積することで行った.
不適切な細胞の除去に関しては,汎用的な細胞培養用のディッシュ内で培養した除去する細胞に対して収束した超音波を付与することで実行した.
細胞シートの回収は,汎用的な細胞培養用のディッシュの培養面全体に接着している細胞に対して,均一な超音波を照射することで実行した.
上記の結果は,マウス由来筋芽細胞株C2C12を用いて示した.筋芽細胞は実際に再生医療に用いられる細胞種である.さらに,超音波が細胞に与える影響を確認するために,細胞の代謝,タンパク質の発現,遺伝子の発現,形態等を確認したが,いずれも異常はみられなかった.このことから,本手法は細胞に対して非侵襲なものであると結論づけた.
