Outline of Annual Research Achievements

本研究は、卵子の受精能やホルモン分泌などの卵巣の潜在的能力 (卵巣予備能) と相関する卵巣内の胞状卵胞数に着目し、卵子発生能との関係を検討した。
これまでの牛の体外発育培養系を用いた研究において、卵巣予備能の指標である胞状卵胞数が多い卵巣 (High 群) に由来する初期胞状卵胞から採取した卵子-卵丘-顆粒層細胞複合体 (OCGC) は、胞状卵胞数が少ない群 (Low群) よりも、卵子発育・成熟に必須なエストラジオール17β (E2) の産生能が高く、卵子成熟能・胚発生能が高かった。また、生体牛でも血中・卵胞液中のE2濃度はHigh群で高い一方、E2産生を促進する働きを持つ卵胞刺激ホルモン (FSH) の血中濃度はLow群で高く、顆粒層細胞のFSH応答性の低下が起きていることが示唆された。そこで、初期胞状卵胞由来の顆粒層細胞を用いて定量PCR法によりFSH受容体などのE2産生に関わる遺伝子の発現量を解析したが、両群間に差異は見られなかった。
上記の結果から、Low群でのFSH応答性の低下の要因を解明するためには、卵胞腔形成前の初期卵胞の発育動態に着目する必要があると考えた。一方、牛において二次卵胞期以前の段階の卵胞を用いた体外発育培養系では、正常な産子はおろか成熟卵子すら得られていない。そこで、体外卵胞発育モデルとして利用可能な培養系の開発を目的として、牛二次卵胞の体外発育培養系の改善に取り組んだ。その結果、牛胎子血清添加培地で培養した場合よりも、血清代替物を添加して培養した場合の方が培養後の卵胞直径は大きくなった。さらに、培養器の設定温度を定法の37℃から、牛の正常な体温である39℃に増加させることで、培養後の卵胞直径が増大した。今後、さらなる培養系の改良に取り組むと共に、卵巣予備能が原始卵胞の活性化と一次卵胞・二次卵胞の発育に及ぼす影響について研究を進める予定である。
