Outline of Annual Research Achievements

ジャイアントベシクル(GV)の自己再生産動態を定常化するうえで,内水相への連続的な基質送達が課題であった.前年度までに開発してきた,マイクロ流体デバイスを基軸とする自動計測システムを用いて発見された,脂質二分子膜を介した濃度勾配に逆らって分子が濃縮される新奇の輸送動態はこの課題の根本的解決につながりうる.この輸送動態がいかなる機構で生じるのかを明らかにすることで,本現象の適用範囲や応用可能性を見定めることができる.本年度はまず,分子濃縮の機構解明を目的として研究を進めた.GVを定常的な流れ環境下にさらすことで生じる膜揺動が本質的な役割を果たすことが分かった.この成果は2020年7月,同年9月,同年11月にそれぞれ開催されたMolecular origin of life,第58回日本生物物理学会年会,細胞を創る研究会13.0にて発表した.これまでGVの流れ環境下での様態を報告した例はあるものの,具体的なGVの機能との連関を議論した報告は限定的であり,これらの成果報告は大きな注目を集め,特に日本生物物理学会年会では,学生発表賞を受賞した.さらに現在,国際誌に原著論文として投稿中である.また,本成果に関連する研究内容について,報告者を連名発表者として,国際学会で招待講演を受けた.
また,開発した自動計測システムの適用範囲を広げる試みとして,前年度から継続してデバイスの表面修飾法の検討を進めていた.結果として,内包効率に優れる油中水滴エマルション遠心沈降法で作成されたリポソームをデバイス内部で計測可能となった.本成果は,2020年7月にACS Omegaにて採択・公開された.
これらの一連の研究成果は国内の関連分野の研究者の注目を集め,複数の共同研究へと発展している.
