Outline of Annual Research Achievements

本研究ではシグマトロピー転位を鍵とする芳香環連結反応の開発に継続的に取り組んできた。本年度は、開発した反応の反応機構の詳細な検討及び、合成的有用性を示す応用例として、機能性分子として注目を集めるらせん状芳香族化合物ヘテロヘリセン類の系統的不斉合成研究を行った。
(1) シグマトロピー転位を鍵とするビアリール合成反応の反応機構解析
以前申請者は、芳香族スルホキシドとフェノールに対してトリフルオロ酢酸無水物を作用させることで、2つの芳香環間に位置選択的に炭素―炭素結合が生成し、ビアリールが得られることを報告した。この炭素―炭素結合は[3,3]シグマトロピー転位により形成すると想定していたが、想定反応機構を支持する積極的根拠は十分とは言えなかった。そこで、申請者は低温下における転位前駆体の観測やその転位過程の追跡、さらには計算化学による各種解析など多面的なアプローチに基づく検討を行い、反応基質の電子的性質と転位段階における遷移状態の構造、反応性の関係性を明らかにした。いまだ理解が十分とはいえないカチオン性硫黄化学種のシグマトロピー転位への性質の理解を深める本成果は、今後の反応開発における指針となりうると考えられる。
(2) [8]ヘテロヘリセンの系統的不斉合成
多彩な反応性を示す硫黄官能基を巧妙に活用した合成経路設計によって、様々なヘテロ原子類(C,O,N,S等)が置換したヘテロヘリセンの系統的不斉合成を達成した。さらに、本合成戦略の長所を生かし、適切な合成手法の欠如から従来困難であったヘテロヘリセン中の置換原子の違いに着目した構造・物性の系統的な評価を行った。その結果、ヘリセンを特徴づけるらせん構造の熱的安定性や、蛍光量子収率、円偏光発光特性など各種物性に対する置換原子の影響は顕著であることが明らかになった。今後、多様なヘテロ芳香族化合物の系統的合成への本戦略の利用が期待される。
