Outline of Final Research Achievements

The Sox transcription factors are thought to play an important role in regulating cell differentiation states by forming various combinations of Sox-partner factor complexes during embryonic development. By inducing genome cleavage by CRISPR-Cas9 followed by DNA repair using a long ssDNA donor, we were able to efficiently knock-in the composite tag sequences into the sox3, sox11a, and pax6a genes.

Academic Significance and Societal Importance of the Research Achievements

CRISPR-Cas9などを用いたゲノム編集による正確なゲノム改変、特に特定のDNA配列の正確な挿入(ノックイン)は効率が低いため、幅広く利用されていない。本研究では、約60アミノ酸からなる複合タグ配列をコードするDNAの正確で効率的なノックインが、長鎖ssDNAドナーを用いることで可能になることを示した。また、タンパク質の機能解析に複数のタグからなる複合タグが有用であることを示した。これらの成果は、タグのノックインを利用することによる遺伝子機能の研究の進展に大きく寄与することが期待できる。
