Outline of Annual Research Achievements

2020年度までに吸入水素が細胞内カルシウム動態に影響を与え、特に小胞体からのカルシウム動員を抑制し、アポトーシスの最終経路であるミトコンドリア孔の開放を抑制効果を有することを発見した。このプレコンディショニング効果は吸入水素が心筋虚血再灌流障害に対して予防効果を有してことを示しており、吸入という簡便で実行可能性のある投与方法も含めて十分に臨床応用可能と考えていた。
2021、2022年度では吸入水素の心筋保護プレコンディショニング効果の作用機序に関するプロトコルを実施していき、細胞内保護カスケードであるMAPK/ERK経路が有意に発現することが確認された。更に、吸入水素やこのMAPK/ERK経路は細胞内のクリーニング作用であるオートファジーを活性化することが示唆されていたため、フローサイトメトリー法により確認したところ、吸入水素によるプレコンディショニング刺激はMAPK/ERK経路の活性化とオートファジー機能の促進の相互的作用を誘導することが認められた。このことは吸入水素がラジカル除去以外に細胞内カスケードを活性化し予防的な心筋保護作用を示唆する内容であり、吸入水素療法の新しい臨床応用を提供するものであった。
2023年度はこの新しい知見に関する文献的考察を重ねて論文作成を行い、年度末にLifeScence誌に論文投稿を行ったところである。

Reason

2020、2021年度のCOVID-19感染症のアウトブレイクで診療応需や入校制限により研究エフォートが減少したことが多大なる影響を及ぼしている。2023年度末に何とか論文投稿を行うことができた。

論文投稿を行った結果、major revisionでwestern blotに関する追加実験の算段を行っている。
