Outline of Final Research Achievements

In the present study, we have developed a technique to make an implant which consists of collagen and human adipose-derived stem cells. First, we mainly examined optimal conditions for cartilage regeneration. Various in vitro experiments suggested that human adipose-derived stem cells should be used after chondrocyte differentiation by chondrogenic cytokines such as BMP-2 and TGF-beta1. These implants could form cartilage tissues in animal transplantation experiments.

Academic Significance and Societal Importance of the Research Achievements

骨、軟骨の欠損は骨折などの外傷や変形性関節症、感染症などにより引き起こされ、治療が困難になる場合も珍しくない。軟骨欠損に対しては、自家軟骨細胞から造形した軟骨組織が保険収載されているが、関節鏡視下での細胞採取の侵襲を要し、また移植に際しては欠損部に合わせた形状の細工を術中に行う必要があり、欠損部に精密に整合させることも困難である。脂肪組織は局所麻酔下に皮下から容易に採取でき、豊富に間葉系幹細胞を含むことが知られている。本研究は低侵襲で簡便に軟骨移植用インプラントを造形することを可能にするものと期待される。
