Outline of Final Research Achievements

(1) BMP-9 directly induces GSK3β phosphorylation in osteoblasts (2) Low-intensity pulsed ultrasound (LIPUS) induces BMP-9-induced osteoblast-like differentiation in human periodontal ligament fibroblasts (hPDLFs) (3) BMP-9-induced osteoblast-like differentiation of hPDLFs was suppressed in an inflammatory environment (in the presence of IL-1β), and the activin A-follistatin system may function as a negative feedback mechanism for BMP-9 in hPDLFs. (4) In two-wall intrabony defects model using beagle dogs, a tendency of periodontal tissue regeneration by BMP-9 was observed.

Academic Significance and Societal Importance of the Research Achievements

本研究より、BMP-9による歯周組織細胞の分化制御メカニズムの一端が示された。BMP-9は結合する受容体やシグナル伝達経路、抑制機構などにおいてBMP-2と異なる特徴を多く持ちながら、強力な骨芽細胞への分化能、骨誘導能を有している。このことからBMP-9はBMP-2と比較してその投与量を低減することによりコスト面や安全性という観点から大きな利点を有していると考えられ、BMP-9の臨床応用に向けたトランスレーショナルリサーチを今後さらに推進していく必要があると考えている。
