Outline of Final Research Achievements

Cellular nonsense-mediated mRNA decay (NMD) degrades mRNAs carrying a premature termination codon (PTC). Readthrough therapy can suppress PTC and generate full-length protein from aberrant mRNAs. NMD activity affect efficiency of the readthrough therapy. Here we investigate whether nutritional status influences the efficiency of readthrough therapy for Duchenne muscular dystrophy. Our results show that high-fat diet feeding potentiated gentamicin-mediated readthrough therapy for the mdx mice, an animal model of the Duchenne muscular dystrophy.

Academic Significance and Societal Importance of the Research Achievements

本研究では、Duchenne型筋ジストロフィーに対するリードスルー治療において、食生活の変化(高脂肪食)が治療効果に影響することを明らかにした。高脂肪食は健康に良い食事とは言えないが、リードスルー治療期間専用の食事としての可能性が示唆された。本研究成果は、ナンセンス型遺伝子変異疾患のリードスルー治療のための栄養基盤の創生につながるものと期待される。
