Outline of Final Research Achievements

PML body is one of nuclear domains and considered to regulate gene expression. However, its role in transcription remains unclear. By developing a new technology “ALaP (APEX mediated Chromatin Labeling and Purification)”, which enables us to identify the interactions between PML body and chromatin, we uncovered that PML body activates transcription of male specific genes.

Academic Significance and Societal Importance of the Research Achievements

これまで、遺伝子の転写制御の研究は、シグナリングや転写因子、エピジェネティクスなどを含むクロマチン構造に焦点をあて行われてきた。しかし、クロマチン構造の解析だけでは、すべての転写制御の仕組みを解明することはできない。本研究によって、転写制御における役割が未解明であった核内ボディによる転写制御メカニズムが明らかになったことで、転写制御分野の更なる発展が期待できる。
