Outline of Final Research Achievements

Genomic DNAs in eukaryotes are tightly packaged in chromatin structures. For DNAs to work, they must be transcribed by an enzyme called RNA polymerase. However, it was not clear how RNA polymerase can read tightly-packed chromatin DNAs. In this study, we have done in-vitro transcription reactions using RNA polymerase and a chromatin template, flash-froze the reaction mixture, and then solved molecular structures of RNA polymerase transcribing a chromatin DNA.

ヒトなどの細胞において、クロマチンの高次構造の影響が遺伝子のON/OFFに与える影響は極めて大きく、細胞の分化、多能性や、がん化などにも深くかかわっている。本研究は、それらの生物学的な背景を考えるにあたって、新たな知見を与えるものである。また、このように複雑で不均一なタンパク質核酸複合体の構造解析は、従来は非現実的と考えられていたものであり、世界に先駆けて構造解析に成功したことは、技術的な面も大きな意義を持つ。
