Outline of Research at the Start

SARS-CoV2に関する研究は、滋賀医科大学第2病理学教室でも実験を行う予定である。当教室ではSARS-CoV2のWuhan株を感染させたカニクイザルのサンプルがすでに保管されている。これらを用いて、肺炎像が見られた個体と見られなかった個体の血液中のPD-1, TIGIT, IFNs及びIL-6の発現量を解析する。
肺炎像が見られた個体で発現が増加していた因子が、増悪化因子の候補となる。因子が絞られたらその因子を抗体によりノックアウトしたハムスターを用いて、実際に増悪化するのか検討する。この研究により、インフルエンザおよび新型コロナウイルス感染症共通の増悪化因子を同定することができる。

Outline of Final Research Achievements

The molecular mechanisms of host immune responses involved in the severity of influenza remain largely unclear, particularly the relationship between severe disease and immunosuppressive factors. The aim of this study is to identify contributing factors to disease severity using cynomolgus macaques infected with H7N9 highly pathogenic avian influenza virus. In macaques that developed severe symptoms, increased expression of immune checkpoint molecules (PD-1 and TIGIT), as well as elevated production of IFNs and IL-6, were observed, suggesting their involvement in disease progression (Suzuki et al., AAC. 2020). Furthermore, we extended this research to analyze SARS-CoV-2 infection, however, severe disease was not observed in cynomolgus macaques.

Academic Significance and Societal Importance of the Research Achievements

今回使用した高病原性H7N9鳥インフルエンザウイルスは、日本の検疫所で摘発された生肉から同定されたウイルス株であり、日本への輸入例はない。H7N9感染カニクイザルでは、一頭が人道的エンドポイントに達するほど重篤な症状を示したことから、人に感染した場合も重篤になることが予想される。今回、PD-1、TIGITやIFNs、IL-6が重症化に関与していたことが明らかとなったことは、今後日本で感染例があった場合に非常に有効な知見となる。
