Outline of Final Research Achievements

We focused on androgen receptor (AR) and its transcriptional role through enhancer activation. As an epigenetic co-factor of AR, we identified that BRD4 played a critical role for prostate carcinogenesis. Furthermore, we identified aberrant enhancer activation, which was independent of AR, in acquisition of castration resistance. We showed that inhibition of histone methyltransferase DOT1L might be a novel therapeutic strategy through regulating aberrant enhancer activation.

Academic Significance and Societal Importance of the Research Achievements

前立腺癌の発癌ならびに去勢抵抗性獲得の分子機構について、エピゲノムの観点から解析を行なった。近年ゲノム解析手法の発達によりゲノムを主軸においた研究が盛んである一方、エピゲノムを主軸に置いた研究は不十分であり、我々の研究によりエピゲノムの観点から新規知見が得られた点で学術的意義が高いと考えられる。また、エピジェネティック阻害による治療戦略の可能性を示した点では、社会的に克服すべき治療抵抗性病態である去勢抵抗性前立腺癌に対する臨床応用へと繋がる、大きな社会的意義を有していると考える。
