Outline of Annual Research Achievements

本研究の目的として、腎癌の肺転移成立において最も重要なのは「細胞集団運動」なのか、あるいは「protrusion形成」なのかそしてそれらを制御するmiRNAについて明らかにすることである。
本年度はゲノム編集技術を用いて、miRNAの親株(転移能:低)へのノックインおよび高転移株でのノックアウトを行った。なお、標的とするmiRNAとして、これまでの研究で親株に比較して発現が増加かつ高転移株共通で発現がみられる複数のmiRNAを選択した。得られた、ノックインおよびノックアウト細胞株について、2.5次元培養および3D培養における表現型について比較解析を行った。その結果、いくつかのmiRNAにおいて、高転移株へのノックアウトでprotrusion形成能が低下することが明らかとなった。今後、動物へ細胞株を腎被膜下へ移植し、その転移能について検討を行う必要がある。
