Outline of Annual Research Achievements

頭蓋縫合早期癒合症は先天的に頭蓋縫合が癒合しており、癒合する縫合により特徴的な異常頭蓋形態を示す疾患である。本疾患の原因遺伝子がいくつか報告されているが、なぜ早期に癒合が生じるのかということに関しては未だ分かっておらず、病的に癒合した縫合を調べてもその原因の特定には至っていなかった。そこで、生理的に自然閉鎖するマウスのposterofrontal suture(ヒトのmetopic suture)に着目して検討を行ったところ、エストロゲンレセプタ-α(ERα)が癒合段階にあるposerofrontao sutureで多く発現しているということを見出した。しかし生理的に癒合するのはこの縫合のみであり、sagital sureuteはposterofrontao sutureと連続しているにもかかわらず、sagittal sutureは縫合しない。
そこでマウス頭蓋骨よりsagital sureuteならびにposterofrontao sutureの縫合間の組織を採取し、これを培養後に遺伝子発現をRT-PCTにて検討したところ、FGFR2、ならびにBMP3がsagital sutureでより多く発現していた。FGFR2遺伝子はApert症候群の原因遺伝子であり、その関連が示唆された。またBMP3は骨形成蛋白質ファミリーのうち、唯一の阻害剤である。すなわちこの発現により縫合が開存していると思われる。本蛋白質とERαとの発現の関連が今後の検討事項である。
