Outline of Final Research Achievements

To efficiently label neurons in the white matter, we modified the recently reported FlashTag technology and labeled different brain regions of developing mouse cerebral cortex. During the investigation, we found a regional difference in the migratory profiles of neurons that is dependent on developmental stage, whereas migrations of neurons in the developing mouse cerebral cortex were thought to progress evenly across different brain regions. Furthermore, to analyze the white matter neurons that are abundant in the human neocortex, we obtained approval of the ethics committee and have started histological analysis of the white matter neurons of human postmortem brains.

Academic Significance and Societal Importance of the Research Achievements

フラッシュ・タグ法を改変した新たな手法を確立したことで、子宮内電気穿孔法よりも簡便で、かつ、より広い範囲で神経細胞をラベルすることができるようになった。また、この手法を用いて、これまで均一であると考えらえていた新皮質における細胞移動が、脳の領域によって、違いが大きいことを見出した。これらの新しい手法や所見は、同じ研究分野の他の研究者にとっても有用で、解析の基盤となる所見であると考えられる。また、白質神経細胞は、ヒトを始めとする霊長類に特徴的であるため、ヒトの脳を用いた解析が必須となる。ヒトの死後脳組織を用いた解析を開始したため、最終的な目的である病態の解明への第一歩が踏み出せた。
