Outline of Final Research Achievements

We have established an experimental system that allows to track cell clonality and clone size using genome barcoding and transposon, and generated mice that carry these constructs. This model will become a platform useful and applicable to other field of research than just hematology.

Academic Significance and Societal Importance of the Research Achievements

造血幹細胞機能に対する造血ストレスの影響は、昨今非常に活発な血液研究分野の一つである。本研究では、炎症反応が起きる場である血液の幹細胞をモデルとして、感染や炎症など日常的に起こる免疫活性化に起因した造血ストレスがクローナル造血の発症・進展に関与するという新たな仮説の検証を行うため、シングルセルからのゲノムバーコード検出を可能にする高感度ゲノムバーコード増幅法やクローンの数とサイズの評価を可能にするゲノムバーコードマウスの作成を行なった。これらの技術や実験材料は、造血幹細胞の分野だけでなく、幹細胞生物学や癌生物など他生物医学研究領域にとっても有用なツールを提供し、次世代医療技術の発展につながる。
