Research Abstract

本特定領域研究は、生きている状態の生体試料中で機能し、各種生体関連分子群の時空間的なデジタル計測を可能とする蛍光プローブ開発を目的とするものである。本年度はまずグルタチオンS-トランスフェラーゼ(GST)活性を検出可能な蛍光プローブの開発を行った。GSTは薬物第二相抱合反応を司る酵素として広く知られているが、近年では他の生体内酵素群の機能調節タンパク質として働いていることも指摘されており、その細胞内動態、活性制御機構に注日が集まっている。プローブは申請者がこれまでに確立してきた光誘起電子移動を原理とする設計法に基づいて設計・合成した。具体的には蛍光個としてフルオレセインを持ち、GST反応部位としてジニトロベンゼン誘導体を有するプローブを合成したところ、プローブ自身はほぼ無蛍光であるが、GSTによりグルタチオン化され強蛍光性の生成物を与えることが示され、GST活性検出蛍光プローブとして機能することが示された。そこで次に本プローブを種々の培養細胞へと適用したところ、ある種の細胞ではWestern blotによってGSTが豊富に存在することがわかっているにもかかわらず、その活性はきわめて低く抑えられていることが明らかとなった。本細胞のLysateを作成すると、そのGST活性は極めて高いため、細胞が生きている状態では何らかの機構によりGSTの活性が制御されていることが明らかとなった。本知見は、生細胞内酵素活性検出蛍光プローブの開発によって初めて得られたものであり、今後本プローブを用いた様々な応用研究が期待される。
