Research Abstract

細胞のシステムとしてのメカニズム解明の鍵を握る性質の1つと考えられるのは、細胞表面の局所的なイオンチャネルや親水性/疎水性などに関連した電位情報である。細胞を生きたまま観察できる原子間力顕微鏡に、表面電位を計測する機能が付加されれば、生体細胞ダイナミクスの本質を探ることができる。本研究の目的は、『探針や細胞表面との間で電荷の授受を行うことなく、細胞の表面電位を高感度・高分解能に画像化すう新しい計測技術を開発すること』である。具体的には、光異性化反応で分極の有無が切り替わるフォトクロミック材料で顕微鏡探針を修飾し、外部からの刺激光により探針の分極を切り替えることにより、細胞表面の電位を計測するという新しい原理に基づく計測技術を開発することをめざす。具体的には、以下の課題について検討した
1. 現有の原子間力顕微鏡において、溶液中に置かれた探針の先端にレーザ光を集光することが困難である。溶液セルの形状を工夫・最適化することにより、効率的にレーザ光を集光できるようにした。また、分極に変調を与えられるように、レーザ光をスイッチング制御できるように制御システムを改良した。
2. Siからなる顕微鏡探針を実際にフォトクロミック材料で修飾して、分極変化による電位変化を検出できるかどうかを検討した。具体的には、試料として導電性試料を取り上げ、探針・試料間に静電気力が作用している状態で制御光を変化させ、探針の分極変化による変調成分が電位として検出させることを確認した。
3. 溶液中において細胞表面の電位を計測できることを明らかにした。
