Research Abstract

生体内において、選択的な酸化反応を行う金属酵素、例えば,ヘム依存性のHorseradish Peroxidase(HRP)は、アミノ酸によるPush-Pull効果によって、ヘテロリシスを誘起することが知られている。この点に着眼し、強いドナー性配位となるアミドアニオン配位を導入した単核鉄錯体を設計、合成した。本錯体は、マイナス40度の低温下、過酸化水素との反応により、単核鉄3価ヒドロペルオキシド種を与えた。安定性を評価した結果、Push効果によって、ヒドロペルオキシド基の酸素-酸素結合のヘテロリシスの誘起されることを見いだした。また、シクロヘキセンを酸化基質とし、触媒酸化反応を検討した結果、ドナー性の高いアミドアニオン配位子を用いるほど、鉄3価ヒドロペルオキシド種の安定性が低下することを見出した。また、中性の配位子を用いた場合のように酸素-酸素結合のホモリシスが優先的に進行するのではなく、テロリシスが誘起されるために、シクロヘキセンのアリル位の水酸化ではなく、エポキシ化生成物が得られることが判明した。この結果は、アミドアニオン配位の導入によるPush効果によって、単核鉄3価ヒドロペルオキシド種の酸素-酸素結合のヘテロリシスが進行し、アリル位の酸化の原因となるヒドロキシルラジカルの生成が抑止されたためと考えられる。
