Research Abstract

モリブデン酵素の活性部位はプテリンコファクターと呼ばれる特徴的なシチオレン骨格を有し、一方の硫黄原子とプテリン間のNHは分子内NH…S水素結合が可能な距離にある。本研究では、この酵素のモデルとして、1,2-ベンゼンジチオラートの一方の硫黄のみに分子内NH・・・S水素結合が形成可能な配位子を新規に設計・合成し、モノオキソモリブデン(IV)錯体を合成した。同定はX線結晶構造解析、NMR、IR、可視紫外吸収スペクトル、元素分析などで行い、水素結合の形成を固体、溶液中で確認した。4価錯体において、水素結合の形成によりMo-S結合は長くなり、酸化還元電位は正側にシフトして観測された。酵素の基質であるトリメチルアミン-N-オキシドの還元反応は、水素結合が無い場合に比べて速く両方の硫黄に水素結合した錯とは、ほぼ同等であることが明らかとなった。生成したジオキソモトブデン(VI)錯体の溶液構造はオキソのトランス位の硫黄へのみNH・・・Sを形成した異性体だけが存在していた。また、ラマンスペクトルの結果は、トランス位の水素結合がMo=O結合を強めていることを示している。即ち、2つのチオラートのうち、オキソのトランス位の水素結合が反応性、安定性に対して支配的であることがわかった。酵素活性部位に於いても、一方の硫黄へのNH…S水素結合が酸素移動反応の活性化制御を行っていること示している。天然では水素結合が蛋白質の構造変化と協奏的に形成・切断されることで、酵素の反応性が巧に制御され、効率のよい触媒サイクルが実現されていると考えられる。
