Research Abstract

アラメチシンは20残基からなる直鎖状ペプチドで、細菌trichoderma virideから得られる抗菌ペプチドである。このペプチドは膜に結合して電位駆動型イオンチャネル活性を持つことが知られている。本研究ではカルボニル炭素の大きな化学シフト異方性に注目して、化学シフトテンソルをペプチドが膜法線の周りで回転している運動モデルを用いて解析した。この結果、N-末端はα-ヘリックスを形成して膜法線から17度傾き、C-末端は3_<10>-ヘリックスを形成して膜法線から32度傾いている詳細な立体構造と膜に対する配向が明らかになった。
バクテリオロドプシンはHalobacterium Salinarumの紫膜に存在する内在性膜タンパク質であり、光駆動型プロトンポンプ活性をもっている。本研究では[ζ-^<15>N]Lys-bRの^<15>NCP-mS-NMRスペクトルを観測して、圧力と光により誘起されるレチナールの異性化を観測した。この結果、圧力順応状態では13-cis状態が増加し、明順応状態ではall-trans状態が増加することが判明した。さらにTyr-185近傍のタンパク質の主鎖は2つの構造が共存したことから、レチナールの異性化は近傍のタンパク質に局所的に影響を与えることが分かった。
グルカゴンについて脂質二分子膜の存在下で線維形成を行ったところ、水溶液中で形成する線維はN-末端側とC-末端側がβ-sheet構造を形成するのに対し、脂質二分子膜存在下では、N-末端はα-helixを形成したままであることが判明した。また脂質二分子膜存在下では水溶液中に比べて核形成速度は速くなり、線維成長速度は遅くなることが分かった。グルカゴンは脂質膜に結合して濃縮されるので線維核形成速度は速くなったことが明らかになった。
