Research Abstract

アデノシン受容体のサブタイプであるA1アデノシン受容体のホモダイマー形成に関与する部位(インターフェイス)の検索をおこなう目的でTM4とTM5に存在する特定のアミノ酸残基を変異させ、BRET法により解析したところ、これらのアミノ酸はダイマー形成には関与せず受容体活性そのものに重要であることを偶然に見いだした。このアミノ酸はGPCR間にて保存性の高い132番目のTrpであり、分子モデリングからもリガンド結合に関与するTM3とTM5の4つの残基と近接しており、リガンド結合活性に重要な役割を果たしていることが推測された。
