Research Abstract

移動細胞の先導端にはラメリポディアとよばれるアクチン骨格を主成分とする突起構造が形成され、細胞運動を制御する生体ナノシステムとして機能する。アクチン脱重合因子であるコフィリンは先導端ラメリポディアに局在し、アクチン繊維の脱重合を促進すると考えられているが、細胞内では、刺激に応答したアクチン重合ならびにラメリポディアの形成と伸長に必要であることが知られている。このような刺激依存的な細胞内アクチン重合の促進機構を解明するため、可逆的な光活性化蛍光蛋白質であるDronpaとアクチンの融合蛋白質の蛍光強度の減衰量の測定による細胞内アクチンモノマーの時空間的変化を生細胞で測定できるFDAP(fluorescence decay after photOactivation)-time lapse imaging系の開発に成功した。本法を用いて、ニューレグリン刺激依存的なラメリポディアの形成過程における細胞内のアクチンモノマー量の経時的変化を測定し、刺激後2-3分後にアクチンモノマーは大きく減少することを見出した。コフィリンを不活性化した細胞では、刺激前のアクチンモノマー量が低く、ラメリポディア形成が阻害された。さらに、ラメリポディア形成細胞における細胞内のアクチンモノマー量の空間的変化の測定にも成功した。本研究で開発したFDAP time lapse imaging法という新しい顕微鏡イメージング技術はアクチンモノマーの時空間的変化を測定できるだけでなく、他の多くの可溶性因子の変化の測定にも応用可能であり、今後広く応用されることが期待される。
