Research Abstract

藍色細菌の時計タンパク質KaiCを生化学的、遺伝学的に解析し、KaiCが極めて微弱なATPase活性を有することを発見した。KaiCはN末端ドメインとC末端ドメインの2つのドメインからなり、共に一セットのATPaseモチーフを持っているので、ATPase活性の存在が推測されていたが、あまりに活性が微弱なため、これまでその存在は証明することができなかった。今回、好熱性藍色細菌Thermosynechococcus elongatus由来のKaiCを極めて高度に精製することに成功し、KaiCの2つのドメインが共に極微弱なATPase活性を有すること明らかにした。この活性が大腸菌由来のATPaseの混入によるものでないことは、ATPaseモチーフにアミノ酸置換変異を導入したKaic変異体ではその活性が完全に消失することで確認した。極めて興味深いことにKaiC ATPase活性は反応温度を変えても変化しなかった(温度非感受性)。生物時計の周期(速度)は温度に依存せず温度補償されているので、時計のこの特性の少なくとも一部はATPase活性の温度非感受性(温度補償性)で説明できるのかも知れない。アミノ酸置換変異を導入してリン酸化サイトをつぶしたKaiC変異体では、この温度非感受性が失われた。
