Research Abstract

ミオシン繊維の動態と繊維化の解析
EGFPミオシンを発現させた細胞で、全反射蛍光顕微鏡によって細胞腹側の細胞膜近傍のミオシン繊維の挙動を高感度カメラにより画像取得し、細胞運動に伴う挙動を解析した。ミオシンの繊維化、脱繊維化がどのように起きるのか、また、個々のミオシン繊維がどのように細胞の尾部や分裂面に集まるかを画像化し解析した。
ミオシン遺伝子の改変による細胞内構築の解析
ミオシンが細胞尾部に集合するのに、ミオシンのモーター活性が必要かどうかを調べるため、ミオシンのモータードメイン内の点突然変異によりモーター活性のないミオシンを形質導入し、細胞尾部に集まるかどうかを調べたところ、モーター活性がなくとも集合できることが分かった。また、ミオシンはリン酸化の制御を受けるので、リン酸化できないようなミオシンで細胞尾部に集まるかどうかを調べた。また、これらの変異ミオシンにGFP標識を行い、細胞内での集合の様子を直接観察したところ、ターンオーバーはリン酸化とモーター活性の両者に依存することが分かった。
ミオシン繊維から細胞の力の出力まで
ミオシン繊維から細胞の力の出力までを観察するために、細胞内のミオシン繊維を観察すると同時に、細胞の出す力を計測した。細胞の出す力を計測する方法としては、最近新しく開発したEvanescence-Force Microscopeを用いた。ミオシンの集合時に確かに細胞の力が発生していることが分かった。
ミオシンの集合の細胞内シグナルの解明 : PTENがミオシンの上位シグナルであることを新たに見いだした。
