Research Abstract

細胞内RNA顆粒は、RNAとRNA結合蛋白、翻訳関連蛋白他の蛋白からなり、遺伝子発現の転写後調節等の制御機能を有している。神経細胞に存在するRNA顆粒は、局所翻訳に関係し、シナプス可塑性の分子的基盤を考える上で重要である。研究代表者らは、RNA結合蛋白(TLS)を含むRNA顆粒が、アクチンモーターであるミオシンVにより、樹状突起からスパインに運ばれることを明らかにした。本研究では、ミオシンV及びTLSの相互作用蛋白を同定し、RNA顆粒の動態を経時的モニター系で解析することにより、神経樹状突起からスパインへのRNA輸送に関する「ナノシステム」の分子制御機構を明らかにすることを目的とする。酵母two-hybrid法により、ミオシンVに結合する蛋白をスクリーニングした結果、12個の候補遺伝子を同定し、海馬初代培養神経細胞系にて解析中である。これらの候補遺伝子のgain-of-function及びRNAiやミオシンVノックアウト(staggerer)マウスを用いたloss-of-functionの実験により、ミオシンVの輸送及びTLSを含むRNA顆粒の輸送との関連を明らかにする。また、網膜神経節細胞においては、TLSはカルシウム依存性にNMDA受容体であるNR1とmyosinVaと結合していることを明らかにした。TLSがカルシウム依存的なNR1スプライシングバリアントの輸送に関与することが示唆された。
