Research Abstract

ショウジョウバエの転写因子Cubitus interruptus(Ci)、およびその動物ホモログであるGlioblastoma(GLI)は、ヘッジホッグシグナル伝達経路上で機能し、形態形成において重要な役割を果たしている。私達は、Ciに結合する因子として、Roadkill(Rdx)を同定し、RdxによるCi活性の制御メカニズムを解析した。Rdxは、Cul3と結合して、ユビキチンE3リガーゼとして、Ciのプロテアソーム依存的分解に関与することが、すでに報告されている。しかし、私達は、Rdxがハエ翅成虫原基の前後軸近辺で、発現が高く、この領域では、RdxかCiの分解に関与しないことを見出した。そして、強いHhシグナルを受け取る前後軸近辺では、Rdxは核膜孔に局在し、Ciとimportinα3との結合を阻害して、Ciの核内移行を阻害することを見出した。一方、比較的弱い強度のHhシグナルを受け取る、さらに前側の領域では、RdxはCiの核内移行を阻害せず、核内のCiの分解を制御する。Rdxの2つの機能の使い分けは、Hhシグナル強度に応じて、核内のCi分子数を調節し、標的遺伝子を選択的に調節するために、重要な役割を果たすことが明らかにされた。この結果は、Rdxが異なるメカニズムによって、転写因子Ciの核内ダイナミクスを緻密に制御しており、この緻密な制御が、正常な形態形成に不可欠の役割を果たしていることを示している。
