Research Abstract

1. アントシアニン分子の修飾ならびに輸送に関与するタンパク質の同定
MYB様転写因子PAP1の過剰発現により発現上昇するアシル転移酵素、カルコン異性化酵素ならびにグルタチオンS-転移酵素(GST)について組換えタンパク質を用いた機能解析とT-DNA挿入ノックアウトラインのフラボノイド分析を行い、蓄積される代謝物変動から機能を解析した。
2. カンプトテシン生産と自己耐性獲得機構について
強力なDNAトポイソメラーゼI阻害剤カンプトテシンを生産する植物の自己耐性機構を調べた。その結果、カンプトテシンを生産する植物では、トポイソメラーゼIに特異的なアミノ酸変異が起こることで、耐性能を獲得していることが明らかになった。また、カンプトテシンを生産しない近縁植物リュウキュウイナモリのDNAトポイソメラーゼIについても調べた結果、生産植物の変異による自己耐性能の獲得はカンプトテシンの生産能の獲得と共進化していることが明らかになった。
3. キノリチジンアルカロイド生合成の細胞内局在と生物学的意義について
Lupinus angusutifoliusにおいてアルカロイド生産するビター品種特異的遺伝子のうち、オルニチン脱炭酸酵素ホモログ(LaDC)、アミン酸化酵素ホモログ(LaAO)、アシル転移酵素ホモログ(LaAT)に関する機能解析と細胞内局在解析を行った。LaDCはリジン脱炭酸活性を有することが明らかになった。さらにキノリチジンアルカロイドを生産する他のマメ科植物も同型の酵素を有することが明らかとなり、本酵素の存在がキノリチジンアルカロイド生産能の分化と関係があることが示唆された。
