液胞と細胞骨格による植物の環境応答および細胞・オルガネラ分化の解析

Research Abstract

本研究では植物の環境適応とオルガネラ分化の研究対象として特に液胞と細胞骨格に着目し、分化・形態形成、細胞分裂周期、環境応答の各過程でのそれらの動態をリアルタイムで観察するとともに、得られた画像情報から独自に開発した立体画像構築プログラムを用いて、空間的かつ定量的解析を行っている。本年度は、細胞分裂、細胞肥大(伸長)の切替えの調節メカニズムを解明するためにカリウムイオン(K^+)を細胞内外に輸送するK^+チャネルの解析を行った。その結果、細胞肥大のみならず、細胞分裂にもK^+チャネルを介したK^+吸収が必要であることが明らかになった。そこで、K^+チャネル遺伝子の発現解析を行ったところ、K^+吸収に関与するK^+チャネル遺伝子NKT1は細胞伸長時でも細胞分裂時でも発現していたが、K^+排出に関与するK^+チャネル遺伝子NTORK1およびNtTPK1は細胞分裂時にのみ発現していた。また、タバコ培養細胞のプロトプラストから液胞を除去したミニプロトプラストの伸長誘導系を確立し、最終的に液胞への水の輸送を担う分子実体である液胞膜型アクアポリン(Nt・TIP)の発現・生理機能解析を行った。その結果、Nt・TIPの過剰発現により液胞の発達と細胞生長が促進され、Nt・TIPが液胞の発達および細胞の体積増加に関与することを明らかにした。一方、シロイヌナズナの気孔開閉運動におけるアクチン繊維の役割について、画像解析により配向、束化、密度などを定量的に評価した結果、気孔開口過程において一過的な束化が生じ、それが解消されることが気孔開口の重要な要件であることを見出した。
