Research Abstract

性特異的な行動の基盤は、中枢回路の発達期に形成される。すなわちエストラジールが視索前野のグルタミン酸依存性スパイン形成を亢進させることがオス性行動発現に重要であると理解されているが、その分子機序は不明である。最近の知見からオス性行動の発現には、視索前野発達段階でのPGE2によるスパイン形成が必須であると考えられた。しかし関与するPGE受容体の種類やその作用機序は不明であった。研究代表者は、8種類のPG受容体欠損マウスを駆使することで、オス性行動の発現に係るEPサブタイプの種類を同定し、さらにPGがグルタミン酸に依存した視索前野スパイン形成を亢進する分子機序を解明することを目指し解析を進めた。その結果、EP4欠損マウスはオス性行動をほとんど示さないことを見出した。またEP4欠損マウスの視索前野初代培養ニューロンでは、PGE2によるフィロポディア形成亢進作用が消失したのみならず、異常形態ニューロンの割合が増大していた。現在、PGE2はこのEP4受容体を介して細胞骨格制御タンパク質の機能を調節することで、先導端ならびにフィロポディア形成を調節するとの仮説に基づき確認実験を進めている。
