Research Abstract

脊椎動物の初期発生過程において、腎臓(中腎)と生殖巣は隣接して形成される。このとき、腎臓を覆う上皮(「腎体腔上皮」とよぶ)と生殖巣は、共に体腔上皮とよばれる同じ上皮組織に由来する。腎体腔上皮が形成される際、上皮細胞としての極性を維持し続けるのに対し、生殖巣の形成過程においては、体腔上皮からの脱上皮化がみられる。本研究では、生殖巣の形成機構を、細胞の上皮-脱上皮(Epithehal-mesenchymal-transition ; EMT)という観点から理解することを目的として解析を進めてきた。具体的には、まず腎体腔上皮に作用させた際にEMTを誘導する遺伝子群を探索し、次にそれらの候補遺伝子に注目して、本来の生殖巣形成のしくみに迫ろうというものである。得られた結果を以下に示す。1)腎体腔上皮に作用させた際にEMTを誘導する能力を有する分子として、Shh,Gli,Ras-MAPキナーゼ因子群などを見いだした。逆に、カドヘリンやCdc42は、上皮極性の維持に必須であった。2)本来の生殖巣の形成過程において、上記分子に注目してその発現や活性化を解析した。その結果、形成初期の生殖巣は、隣接組織(内胚葉)に発現するShhを受け取ることによって、MAPキナーゼ経路を活性化きせることがわかった。これらの分子カスケードが生殖巣細胞にみられるEMTを惹起すると考えられる。3)腎体腔上皮を支持する組織を人工的に除去すると積極的な脱上皮化がみられたことから、正常発生における腎体腔上皮と生殖巣にみられる上皮化の違いは、隣接する支持組織の違い(細胞外マトリックスなど)に起因すると考えられた。
本研究により、初期生殖巣の形成機構について、組織間相互作用、細胞の極性変化、そしてそれらを制御する分子カスケードなどの視点から解明することができた。
