Research Abstract

ヘルペスウイルスはプロテインキナーゼ(PK)をコードしており、宿主細胞因子やウイルス因子をリン酸化し、その活性を制御することによって効率的なウイルスの増殖・生存および病原性に関与している。また、ウイルスPKはウイルス特異酵素であるので、抗ウイルス剤開発の理想的な標的でもある。本研究では、申請者らが世界に先駈けて確立したヘルペスウイルスPKの試験管内アッセイ系を利用して、ウイルスPKの標的宿主細胞因子および標的ウイルス因子を網羅的にスクリーニングすることによって同定し、それらリン酸化の感染細胞における生物学的意義を解析する。これらの解析を通じてウイルスPKによる宿主細胞制御機構およびそれに基づくウイルス増殖調節機構の全体像を明らかにすることを目的と。本年度は、以下の結果を得た。
(i)単純ヘルペスウイルス(HSV)がコードするPKUs3の新規基質としてエンベロープ糖蛋白質gBを同定した。
(ii)gBのUs3によるリン酸化部位として887番目のスレオニンを同定した。
(iii)gBのリン酸化部位の変異体を用いた解析より、Us3によるリン酸化によってgBの細胞表面の発現量が制御されていることが明らかになった。Us3の自己リン酸化は感染細胞の形態を制御していた。
以上、Us3PKの機能発現機構の一端を解明し、Us3の新規機能としてメンブレントラフィックを制御していることが明らかになった。
