Research Abstract

我々はニパウイルスの強力な解析手段となるreverse genetics系を世界に先駆けて開発し、現在でも遺伝子改変ニパウイルスを作出できるのは世界で我々のみである。本研究は、この技術を用いてニパウイルスの病原性発現機序を解析することを大きな目標とし、本年度は主に病原性に最も関与することが疑われている3種類のアクセサリー蛋白に注目して、それらの病原性への関与の機序の解析を行なった。3種のアクセサリー蛋白について、in vitroでIFN応答の阻害作用を持つことをリポーターアッセイによって確認した。そこで昨年度、reverse geneticsによって作出したそれぞれのアクセサリー蛋白を欠損する組換えニパウイルスを細胞および動物に感染させて、病原性への影響およびIFN応答性への影響を解析した。その結果、IFN応答性への影響が減少する傾向が認められアクセサリー蛋白によっては感染細胞中でも作用を及ぼしている可能性が示唆された。そこでこれらウイルスをそれぞれハムスターに感染させて病原性への関与を、臨床症状、生死、病理学的解析により検索したところ、2種類のアクセサリー蛋白欠損ウイルスで病原性の著しい減弱が認められた。現在詳細な機序を解析中である。本研究成果はニパウイルスの病原性に関与するウイルス蛋白を動物実験で証明する初めての成果となると考えられる。
