Research Abstract

腸管感染病原菌は生体から種々な影響を受けていると考えられるが、具体的な因子については十分解明されていない。前年度には、胃酸耐性能とバイオフィルム形成能が相関し、神奈川現象陰性菌に比べて神奈川現象陽性腸炎ビブリオにこれらの活性が強い(高い)ことが明らかになった。本年度は、可能性のある各種の供試因子のうち、血清の関与が疑われた。そこで、血清が腸管感染病原菌の増殖にどのような影響を与えるかについて詳細に検討した。まず、血清中にある細菌に対する抑制因子と促進因子が別の因子であり、各々が単一の分子であるか否かをSephadex 25ゲルろ過することで調べたところ、抑制因子はボイド部分に溶出されたのに対し、促進因子活性は1.5〜1.7ボイド付近に活性の単一のピークとして認め、単一の分子が各々の生物活性を発揮していると考えられた。まず、抑制因子の精製を進めたところ、トランスフェリン(アポ型)が増殖抑制因子として働いていることが分かった。一方、増殖促進因子は完全な精製には至らなかったが、MALDI-TOFMS,LC-MS/MS,FT-IRなどを用いた解析をしたこところ、グリコリピッドであることが分かった。分子量は約2,000、100℃で30分間処理に耐える新奇な生体由来分子であると考えられた。本分子は、腸炎ビブリオのみならずグラム陽性(黄色ブドウ球菌)、陰性菌(ブルニフィカスなど)に広く増殖促進因子として作用することも明らかになった。
