Research Abstract

【目的】主にT細胞に発現されるTNFファミリーCD30LとそのレセプターCD30はT細胞の機能分化や増殖促進、維持に関与しているとか考えられる。Mycobacterium bovis BCGに対する感染防御機構におけるCD30L/CD30の役割を調べるため、CD30LおよびCD30ノックアト(KO)マウスを用いてBCG経気道および腹腔感染実験をおこなった。
【方法】BALB/cおよびC57BL/6背景CD30LKOマウスにBCG(東京株)を経気道または腹腔投与し、感染後14、21、63日目の腹腔内、脾臓、肺の臓器内菌数を測定した。また経時的に抗原特異的CD4T細胞およびCD8T細胞の推移を追うためにAg85または卵白アルブミン(OVA)を発現するBCG株(BCG-OVA)を用い、フローサイトメーターを用いてリンパ球表面マーカー、Ag85、OVA抗原特異的な細胞内IFN-γ産生およびMHCテトラマー陽性細胞の解析を行った。細胞増殖及びアポトーシス感受性の測定を行った。
【結果】腹腔内、肝臓、脾臓、肺の臓器内菌数は、感染早期の7、14日後では、コントロールマウスと顕著な差を認めなかったが、28日目以降では、CD30LKOマウスで肺または脾臓で有意に増加していた。PPDまたはAg85由来のpeptide25特異的CD4+T細胞の産生は、感染28日目以降でKOマウスで著明に減少していた。CD30LのBCG感染後の発現を調べると主にCD4T細胞に発現していた。BCG感染CD30LKOマウス由来のCD4T細胞はCD30L発現P815やCD30L発現T細胞の存在下で抗原刺激によるIFN-γの産生が回復した。
【結論】以上の結果からCD30L/CD30は抗原特異的CD4+T細胞のTh1分化の促進に重要であることが明らかとなった。
