Research Abstract

IL-27はIL-12サイトカインファミリーに属するサイトカインであり、Th1誘導作用に加えて免疫抑制作用を有する。IL-27の持つ免疫抑制作用を主として樹状細胞に関して検討したところ、IL-27受容体を欠損するマウス由来の樹状細胞ではその抑制作用が失われるため抗原提示能が亢進し、抗原でパルスしたこの樹状細胞を個体内に移入することにより腫瘍に対する強い防御免疫を誘導することが出来た。このことから、IL-27シグナルを制御することにより、免疫を増強するアジュバントとしての応用が可能であると考えられ、既に報告した原虫感染の系における有用性をさらに検証している。また、このIL-27シグナルによる免疫・炎症抑制作用を、接触型過敏反応の系において確認し、IL-27の治療効果を見いだした。一方、IL-27受容体欠損は、喘息モデルにおいては症状増悪を来したが、同じI型アレルギー疾患である鼻炎モデルでは逆に症状の軽減を認め、IL-27が異なる疾患・臓器において異なる作用を示すことを見いだした。このことは、IL-27の持つ免疫抑制作用が、感染時などに生じる組織破壊を伴う炎症やサイトカインの過剰産生による自己損傷、ショック病態などに対して治療効果があることを示しており、IL-27の持つ免疫抑制作用を用いた治療応用の可能性を検討している。IL-27は、炎症誘導作用の強いIL-17産生細胞であるTh17の分化を抑制することにより免疫抑制作用を示すとされているが、IL-17には炎症を引き起こすことにより原虫感染に対する免疫誘導作用があることを見いだし、IL-17欠損マウスにおける原虫(trypanosoma cruzi)感染の詳細を解析している。
