Research Abstract

我々はこれまで、FAEのトランスクリプトーム解析データを手がかりに、M細胞に発現する病原体取り込み受容体の探索を行い、2種のGPIアンカー型蛋白Glycoprotein 2(GP2)およびCellular prion protein(PrP^c)を同定した。昨年の班会議において、GP2は、Escherichia coliやSalmonella TyphimuriumなどのI型線毛と選択的に結合し、これらの細菌のM細胞内への取り込みを促進することを報告した。今年度は引き続き、GP2を介した細菌取り込みの生物学的重要性を検討した。その結果、GP2欠損マウスでは、S. TyphimuriumやE. coliのM細胞への取り込みが大きく減少することが分かった。さらに、破傷風毒素フラグメントCを発現する遺伝子改変サルモネラ菌(ToxC-Salmonella)経口免疫モデルにおいて、パイエル板局所における免疫応答を測定した結果、野生型マウスに比べてGP2欠損マウスでは抗原特異的なT細胞の増殖がほとんど認められなかった。以上の結果から、GP2は、I型線毛を持った細菌のパイエル板への取り込みにおいて必要不可欠であるのみならず、その後の粘膜免疫応答を発動においても重要な役割を果たすことが明らかとなった。
一方、内因性プリオンタンパク質であるPrP^cも、GP2と同様にM細胞の管腔側細胞膜に発現しており、抗原取り込み受容体として機能することを明らかとした。
