Research Abstract

新規なエネルギー転換酵素であるH^+-ピロホスファターゼ(H^+-PPase)は、植物液胞膜および原生動物細胞の内膜系、古細菌細胞膜に局在し、高分子合成の副産物として供給されるピロリン酸(PPi)を加水分解して得られるエネルギーを利用し、プロトンを能動的に膜輸送する。既知のF型、V型、P型ATPaseとは一次構造、基質ともに全く異なる新しいイオンポンプである。この分子について、構造を基盤とする作動機構の解明を進め、次の知見を得た。(1)N端およびC端領域は,生物種により配列も長さも大きく異なる。N端に欠落は酵素機能を完全に失わせ、C端側の親水性領域は一部を欠失しても酵素機能を維持できる。(2)プロテオミクス解析により検出されるリン酸化アミノ酸が機能調節に関わるか否かを検討したところ、リン酸化されている可能性はほとんどないこと、またリン酸化アミノ酸を模擬するアミノ酸への置換によっても、活性への影響は見られず、現時点では、リン酸化による酵素機能の調節はないと判定した。(3)数千の変異型H^+-PPaseの中から,基質加水分解活性に比してプロトン輸送活性が高い変異型酵素を得た。基質への親和性は変化せず、H^+/PPi比が高い作動様式を示すものと判断された。これらの知見に加えて、(4)H^+-PPaseを利用してPPi濃度を測定する技術基盤も検討し、組織抽出液をサンプルとした場合であっても、数μM程度のPPiを検出可能な方法を開発した。
